Anatoxin-a(s): Natural Organophosphorus Anticholinesterase Agent
نویسندگان
چکیده
Anatoxin-a(s) is a guanidinemethyl phosphate ester isolated from the freshwater cyanobacterium (bluegreen algae) Anabaena flos-aquae strain NRC 525–17. Previous work has shown anatoxin-a(s) to be a potent irreversible inhibitor of electric eel acetylcholinesterase (AChE, EC 3.1.1.7). Anatoxin-a(s) has been shown to be an active site-directed inhibitor of AChE, which is resistant to reactivation by oximes because of the enzyme-oxime adduct formation. In vivo pretreatment with physostigmine and high concentrations of pyridine 2-aldoxime methochloride (2-PAM) were the only effective antagonists against a lethal dose of anatoxin-a(s). Anatoxin-a(s) is very toxic and it is produced by cyanobacteria during its blooms. Purified toxin has an LD50 (i.p) of approximately 20-50 μg/kg body weight in mice. Toxicoses associated with cholinesterase-inhibiting anatoxin-a(s) have been observed in humans, animals, birds and fish. Anatoxina(s) induces clinical signs of hypercholinergic preponderance, such as salivation, lacrimation, urinary incontinence, defecation, convulsion, fasciculation, and respiratory arrest.

Plasma concentrations of the oxime Pralidoxime Mesylate (P2S) after repeated oral and intramuscular administration.
The use of the oxime P2S as intravenous therapy for organophosphorus anticholinesterase poisoning is well known. In emergency situations this route of administration may prove impractical due to severe symptoms of anticholinesterase poisoning and therefore the intramuscular route is to be preferred. The absolute intramuscular dose of P2S per man, recommended as necessary for adequate therapy of...

This work presents the effects of an anatoxin-a(s) containing extract on a cockroach semi-isolated heart preparation and the results supporting the extract’s biological activity on acetylcholinesterase (purifi ed from eel). The presence of the toxin in cyanobacterial strains Anabaena spiroides (ITEP-024, ITEP-025 and ITEP-026) isolated from the Tapacurá reservoir in Pernambuco, Brazil, was conf...
متن کاملMechanism of action of organophosphorus and carbamate insecticides.
Organophosphorus and carbamate insecticides are toxic to insects and mammals by virtue of their ability to inactivate the enzyme acetylcholinesterase. This review addresses the mechanism of inhibition of acetylcholinesterase by organophosphorus and carbamate esters, focusing on structural requirements necessary for anticholinesterase activity. The inhibition of acetylcholinesterase by these com...
متن کاملAssessment of Anticholinesterase Toxicity, Oxidative Stress and Antioxidant Status in Carbamate and Organophosphorus Pesticides- Exposed Agricultural Workers
Among the numerous pesticides, anticholinesterase compounds are widely used. Their toxicity induced by cholinesterase inhibition at the synapses and neuromuscular junctions, leading to neurological disorders. Inhibition of acetylcholinesterase (AChE) and butyrylcholinesterase (BuChE) has been used as sensitive biomarkers for pesticides exposure. In the present study, AChE and BuChE levels were ...
متن کاملNew method for retrospective detection of exposure to organophosphorus anticholinesterases: application to alleged sarin victims of Japanese terrorists.
With regard to detection of exposure to anticholinesterase, the presently used methods have the disadvantage that they cannot detect either low-level exposures with certainty or the structure of the agent and the extent of poisoning. In principle, organophosphate-inhibited butyrylcholinesterase in human plasma is the most persistent and abundant source for biomonitoring of exposure to organopho...
